摘 要:目的:观察新疆两色金鸡菊醇提物对自发性高血压大鼠(SHR)血压、心肌重构和血浆内皮素(ET)、血管紧张素(Ang)Ⅱ和降钙素基因相关肽(CGRP)含量的影响。方法:将SHR分为模型组、两色金鸡菊醇提物高、中、低剂量组(3.2,1.6,0.8 g·kg-1)、卡托普利组(4 mg·kg-1)及杜仲平压片组(187.5 mg·kg-1),连续ig给药4周。采用无创血压计观察其对血压的影响;计算各组大鼠左心室质量指数(LVWI)和心脏指数;采用放射免疫法测量血浆中ET,AngⅡ,CGRP含量的变化。结果:干预4周后,各给药组大鼠血压均明显下降(P<0.01);与模型组相比各组大鼠LVWI均下降,其中两色金鸡菊醇提物高剂量组和卡托普利组的下降存在显著差异(P<0.01),且高剂量组和卡托普利组心脏指数降低(P<0.05);与模型组相比,两色金鸡菊醇提物高、中剂量和卡托普利可明显减少血浆中ET和AngⅡ的含量(P<0.01),同时血浆中CGRP的含量有增加的趋势。结论:两色金鸡菊醇提物具有一定的降压作用,可以改善SHR心室肥厚情况,其作用机制与抑制体内肾素-血管紧张素系统和改善内皮功能有关。Objective:To observe the effects of the alcohol extract from Coreopsis tinctoria on blood pressure,the content of plasma endothelin (ET),angiotension Ⅱ (Ang Ⅱ) and calcitonin gene-related peptide (CGRP) in spontaneous hypertension rats (SHR).Method:The spontaneous hypertension rats (SHR) were randomly divided into six groups,untreated control,high,middle,low dosage group of the alcohol extract from C.tinctoria (3.2,1.6,0.8 g· kg-1),captopril group (4 mg·kg-1) and du zhong tablet group (187.5 mg· kg-1).Tail blood pressure were measured every week.After 4 weeks,the contents of plasma ET,Ang Ⅱ and CGRP were detected by radioimmunity method respectively.The left ventricular weight index (LVWI) and cardiac index were monitored too.Result:Compared with model group,the blood pressure of all groups significantly lowered after 4 weeks (P <0.01).The LVWI of all groups was decreased,while the high dosage group and captopril group showed significant difference (P < 0.01).The contents of ET and Ang Ⅱ were decreased greatly (P < 0.01) in the high,middle dosage group and captopril group,while the contents of plasma CGRP were increased in all group,but showed no significant difference.Conclusion:The alcohol extract from C.tinctoria could significantly reduce blood pressure and improve myocardial hypertrophy in SHR,which might be related to inhibiting RAS and improving the endothelial function.
