摘 要:目的观察潜阳合剂(QYHJ)对自发性高血压大鼠(SHR)胸主动脉血管重构的影响。方法将12周龄SHR随机分为QYHJ高、中、低剂量以及福辛普利组、SHR对照组,每组8只;另以8只同周龄雄性WKY大鼠为正常对照组。QYHJ高、中、低剂量组给药剂量分别为22.94g/kg、11.47g/kg和5.74g/kg,福辛普利组给药剂量为0.9mg/kg,连续给药8周。SHR对照组和正常对照组给予等容积双蒸水。至20周龄时采集胸主动脉标本,分别行苏木素伊红染色和Masson染色,测量各组大鼠胸主动脉重构指标。结果 SHR对照组胸主动脉血管外径(ED)、内径(LD)、中膜厚度(MT)、中膜厚度与内径比(MT/LD)、血管中膜横截面积(MCSA)、血管横截面积(CSA)、血管中膜横截面积与血管横截面积比(MCSA/CSA)、血管中膜胶原容积分数(CVF)值均较正常对照组增加(P<0.05或P<0.001),发生明显肥厚性重构。QYHJ高剂量组大鼠胸主动脉ED、LD、MCSA、CSA值较SHR对照组减小(P<0.05或P<0.01);MT值较SHR对照组有减小趋势,但差异无统计学意义;MT/LD、MCSA/CSA值与SHR对照组差异无统计学意义。SHR对照组胸主动脉血管中膜CVF值均较WKY大鼠增加(P<0.001);QYHJ高、中剂量组大鼠胸主动脉血管中膜CVF均较SHR对照组降低(P<0.05或P<0.01)。结论 QYHJ对SHR胸主动脉肥厚性重构具有一定改善作用。Objective To observe the effects of Qianyang Heji decoction (QYHJ)on the arterial remodeling of the thoracic aorta in spontaneously hypertensive rats(SHR). Methods Forty 12 week - old male SHR were assigned to 5 groups randomly:SHR control group, SHR+ Fosinopril, SHR-- QYHJ High, SHR+ QYHJ Medium, SHR; QYHJ low. Each group consisted of 8 animals. 8 male WKY rats (12 week- old) were used as normal control group. SHR+ QYHJ High, Medium and Low were given QYHJ contained crude drug of 22.94 g/kg,ll. 47 g/kg,5.74 g/kg respectively; SHR+Fosinopril were given suspl of fosinopril of 0.9 mg/kg; SHR and WKY control group were given the distilled water of the same volume. Drugs were given for 8 weeks constantly. Thoracic aorta was harvested at 20 week- old. After hematoxylin and eosin staining, Masson' s trichrome staining, the values of arterial remodeling of the thoracic aorta of each group rats were measured. Results The values of ED,LD,MT,MT/LD,MCSA,CSA,MCSA/CSA of tho- racic aorta of SHR were increased compared with that of WKY (P;0.05 or P;0. 001). Hypertrophic remodeling was developed on the thoracic aorta of SHR. The values of ED,LD,MCSA,CSA of SHR+QYHJ High were reduced compared with that of SHR (P〈0.05 or P〈0.01). The values of MT of SHR+QYHJ High were reduced compared with that of SHR, but there was no statistic sig- nificance. The values of MT/LD and MCSA/CSA of SHR were not significantly different from that of three groups of SHR+QYHJ. The values of CVF of SHR were increased compared with that of WKY (P〈0. 001). The values of CVF of SHR+QYHJ High and Medium were reduced compared with that of SHR (P〈0.05 or P〈0.01). Conclusion QYHJ could improve hypertrophic remode- ling of thoracic aorta in SHR.
