摘 要:以冬小麦东农冬麦1号为试验材料,将三叶期麦苗经4℃冷胁迫处理7 d后提取叶片蛋白(25℃处理为对照组),利用双向电泳(2-DE)技术分析差异表达蛋白。所得双向电泳图谱经ImageMaster 2D Platinum 7软件分析检测到26个差异表达蛋白质点,选取其中3个进行质谱(MALDI-TOF-MS)分析,所得肽段序列经数据库搜索比对获得的匹配蛋白有3-磷酸甘油醛脱氢酶、ATP酶和Rubisco活化酶。这3种蛋白可能通过提高植物的光合能力、促进植物体内糖分的积累、改善植物对水合离子吸收等方面参与植物的抗冷机制。Total proteins from the leaves of winter wheat Dongnongdongmai 1 were extracted when seedlings at three-leaf period treated at 4 ℃ cold stress for seven days (seedlings treated at 25 ℃ as con- trol), and differentially expressed proteins were analyzed by two-dimensional electrophoresis (2-DE). The 2-DE maps obtained were analyzed by ImageMaster 2D Platinum 7 software and 26 differentially expressed protein spots were detected, 3 spots were selected for MALDI-TOF-MS analysis and peptide fragment se- quences were searched in the database which were matched to glyceraldehyde-3-phosphate dehydroge- nase, ATPase and Rubisco activase. These three proteins might involved in the mechanisms of cold resis- tance through enhancing the photosynthetic capacity, promoting the accumulation of sugar contents and im- proving the absorption of hydrated ion in plants and so on.
