摘 要:目的 :研究肥胖、胰岛素抵抗与候选基因 PC- 1之间的关系及其在中国人群分布的规律。方法 :运用聚合酶链反应 -限制性片段长度多态性 (polymeras chain reaction- restriction fragment length polymorphism,PCR- RFL P) ,分析了 5 3例正常对照者 ,2 7例单纯肥胖者 (OB) ,33例肥胖伴高血压、高血脂症者 (OB+HL) ,35例肥胖伴高血压、高胰岛素血症者 (OB+HI)的浆细胞膜糖蛋白 (PC- 1)基因第 4外显子 12 1位谷氨酰胺 /赖氨酸多态性 (K、Q等位基因 )。结果 :Q等位基因频率在正常对照、肥胖人群、非肥胖糖尿病人群、糖尿病伴肥胖人群分别为 3%、18%、4%和 30 %。与正常对照组比较 ,OB、OB+HL、OB+HI人群的相对危险性 (RR)分别为 4.2 6 ,6 .35 ,8.6 2。与 OB组比较 ,OB+HL、OB+HI人群的 RR分别为 1.33和 1.81。结论 :PC- 1Q等位基因与肥胖、胰岛素抵抗的发生呈显著性正相关。 OB+HL、OB+HI者比 OB者具有更高的 PC- 1Q等位基因频率。它参与了部分中国人肥胖的发生。Objective[WT5,5”BZ]:To evaluate the relationship between the plasma membrane glycoprotein (PC 1) gene and obesity in Chinese people. Methods:The fifty three non obese, healthy individuals, 27 patients with the simple obesity, 33 patients with the obesity associated with hypertension and hyperlipoidemia patients, 35 patients with the obesity associated with hypertension and hyperinsulinemia patients were detected with the technique of PCR RFLP in glutamine for lysine 121 (Q and K allele) polymorphism in exon 4 of PC 1 gene. Results:The frequency of PC 1 gene Q allele was 3%, 18%, 4% and 30% in the controls, obese subjects, diabetic patients and obese diabetic individuals, respectively. Compared with the control group, the relative risk (RR) in simple obesity group, obesity with hypertension and hyperlipoidemia (OBHL) group, obesity with hypertension and hyperinsulinemia (OBHI) groups was 4.26,6 35 and 8.62 respectively. Compared with the simple obesity group, the RR in OBHL and OBHI groups was 1 33 and 1 81 respectively. Conclusion:The frequency of PC 1 gene Q allele are strongly correlated to the obesity and related metabolic disorders in Chinese. The results suggest that the PC 1 gene Q allele is a genetic marker susceptible to obesity. [
