摘 要:目的:研究合欢花黄酮对精神分裂症模型大鼠学习认知能力的干预作用并探讨其作用机制。方法:合欢花黄酮预处理大鼠4周后,腹腔注射地卓西平马来酸盐构建精神分裂症模型,于模型建立后进行Morris水迷宫实验观察各组大鼠的学习认知能力。水迷宫试验后取脑组织,利用免疫组化和免疫荧光观察各组模型大鼠海马组织中S100-β、c-Fos蛋白中的表达定位情况,同时利用Western blot检测海马组织内S100-β、c-Fos蛋白的表达变化情况。结果:合欢花黄酮干预组大鼠的学习认知能力较精神分裂症模型组大鼠有明显的提升(P<0.05)。免疫组化和免疫荧光结果显示模型组大鼠海马组织中S100-β、c-Fos蛋白表达增多,而合欢花黄酮干预后其表达减少(P<0.01)。Western blot结果显示合欢花黄酮能够降低模型组大鼠海马组织中S100-β、c-Fos蛋白表达(P<0.01)。结论:合欢花黄酮能够改善精神分裂症模型大鼠的学习认知能力,并且与海马组织中S100-β、c-Fos蛋白表达变化相关。Objective:To investigate the effect of Flos Albiziae flavonoids on learning and cognitive ability in schizophrenia's model rats and the mechanisms. Methods: Rats pretreated with Flos Albiziae flavonoids for 4 weeks were intraperitoneally injected with dizocilpine maleate to establish schizophrenia's models. Morris water maze test was performed to determine the learning and cognitive ability of rats in different groups after the model establishment. The brains were taken out after the water maze test and the expressions of SIO0-I3 and c-Fos in rat hippocampus were detected by immunohistochemical staining, immunofluorescence, and Western blot, respectively. Results: Learning and cognitive abilitY in model rats treated with Flos Albiziae flavonoids were significantly improved compared with that in model rats without treatment (P 〈 0.05 ). As immunohistochemical staining and immunofluorescence shown, the expressions of S100-β and c-Fos in model rats hippocampus were increased, whereas decreased after treatment with Flos Albiziae flavonoids (P 〈 0. 01 ). The result of Western blot also showed that Flos Albiziae flavonoids reduced the expressions of S100-β and c-Fos in model rats (P 〈 0. 01 ). Conclusion: Flos Albiziae flavonoids can improve the learning and cognitive ability of schizophrenia' s model rats and this action is related with the chanze of S100-β and c-Fos expressions in the hiunocampus.
