摘  要:目的研究褪黑素对宫内缺血缺氧胎鼠脑组织核因子κB(NF-κB)、B淋巴细胞瘤/白血病2基因(Bcl-2)和凋亡相关基因(Bax)蛋白表达的影响。方法 75只妊娠19dSD大鼠均分为空白对照(BC组)、胎鼠宫内窘迫模型对照(MC组)和褪黑素处理(MT组)三组。BC组实施假手术,MC组和MT组建立胎鼠宫内窘迫模型;MT组同步腹腔注射褪黑素10mg/kg。每组按再灌注1、6、12、24和48h均分为5个亚组,分别于相应时间点剖宫取脑组织行HE染色,用免疫组化方法检测NF-κB、Bcl-2和Bax蛋白表达。结果 MT组在再灌注12、24和48h时NF-κB及Bax蛋白表达明显低于MC组,Bcl-2蛋白表达在12h时高于MC组(P<0.05或P<0.01),24h时Bcl-2/Bax比值升高(P<0.01)。结论腹腔注射褪黑素在凋亡调控基因水平对宫内缺氧胎鼠脑损伤有保护作用。Objective To study the influence of melatonin on the expressions of NF-κB, Bcb2 and Bax protein in the brain of fetal rats with intrauterine ischemia and hypoxi Methods A total of 75 rats with gestation for 19 days was equally assigned into 3 groups of BC(blank control), MC(model control) and MT(treated with melatonin). The rats in group BC were shame operated. The rat fetal intrauterine distress model was established in groups of MC and MT, and melatonin 10 mg/kg was intraperitoneally injected at the same time. The rats of each group were subdevided into 5 groups according to repeffusion times of 1,6, 12,24 and 48 h, at which the fetal rats were decapitated and brain tissues were collected for observing pathological changes by HE staining and detecting the expressions of nuclear factor kappa B(NF-xB) ,B cell lymphoma/leukemia-2(Bcl-2) and Bcl-associated X protein(Bax) in cerebral cortex. Results Compared to group MC, in group MT, the expressions of NF-κB and Bax protein at 12,24 and 48 h of reperfusion were lower, the expression of Bcl-2 protein at 12 h of reperfusion was higher(P〈0. 05 or P〈0. 01), and Bcl-2/Bax ratio was increased at 24 h of reperfusion(P〈0. 01). Conclusion On the level of apoptosis regulation gene,intraperitoneal injection of melatonin has a protective effect on intrauterine ischemia and hypoxia-induced cerebral injury.
