摘 要:目的探讨赖氨大黄酸(rhein lysinate,RHL)对2型糖尿病小鼠胰腺保护作用。方法建立小鼠2型糖尿病模型并根据体质量分层,采用随机数字表分为3组:模型组、25 mg/kg RHL和50 mg/kg RHL治疗组,另外设对照组为同龄正常小鼠。检测体质量、血糖、血脂、胰岛素水平;检测胰腺活性氧水平和抗氧化酶活性。体外培养小鼠胰岛β细胞系,检测细胞凋亡情况。结果与对照组相比,糖尿病模型组小鼠血糖、总胆固醇、甘油三酯和胰腺活性氧水平均显著升高(P<0.05),体质量、胰岛素和抗氧化酶活性均显著降低(P<0.05)。与模型组相比,25、50 mg/kg RHL治疗组血糖、总胆固醇、甘油三酯和胰腺活性氧水平均显著降低(P<0.05),体质量、胰岛素和抗氧化酶活性均显著升高(P<0.05)。体外研究发现RHL能抑制过氧化氢诱导的胰岛β细胞凋亡。结论 RHL对2型糖尿病小鼠胰腺的保护作用与抑制氧化应激、减少胰岛β细胞凋亡有关。Objective To investigate the protective effect of rhein lysinate (RHL) to pancreas in type 2 diabetic mice. Methods Mouse model of type 2 diabetic was established, and then were stratified according to body weight. They were divided into the following groups by random number table, model, 25 and 50 mg/kg RHL treatment groups. Another 10 normal mice were designated as normal control. Body weight, blood glucose, total cholesterol, total triglyeeride, insulin, level of reactive oxygen species and activities of anti-oxidation enzymes were determined in 4 weeks after treatment. After pancreatic beta-cell line NIT-1 were treated by RHL at 0, 5 and 10μmol/L for 24 h, cell apoptosis and the expression of Bel-2 and Bax were detected by flow cytometry and Western blotting. Results In comparison with normal control group, blood glucose, total cholesterol, triglyceride and the level of reactive oxygen species of pancreatic tissues were increased, however, insulin, body weight and the activities of anti-oxidation enzymes were decreased in the model group ( all P 〈 0.05). RHL treatment resulted in significant decreases in blood glucose, total cholesterol, triglyceride and the level of reactive oxygen species in pancreatic tissues, while obvious increases in insulin, body weight, and the activities of anti-oxidation enzymes when compared with model group ( all P 〈 0.05 ). In vitro study showed that RHL inhibited the apoptosis in NIT-1 cells induced by hydrogen peroxide. Conclusion RHL protects the pancreas in type 2 diabetic mice, which might be correlated with its inhibiting oxidative stress and beta-cell apoptosis.
