摘 要:[目的]研究瓦尼木层孔菌WN-3分类地位的分子生物学验证及生理学特性。[方法]采用分子生物学方法对瓦尼木层孔菌WN-3菌株进行分类地位验证,通过生长速率法研究该菌株的生理学特性。[结果]菌株WN-3的ITS序列长度为756 bp,在Genbank核酸序列数据库中比对,试验菌株与瓦尼木层孔菌的相似率为98%,确定菌株WN-3为瓦尼木层孔菌,GenBank登陆号为HQ845057。瓦尼木层孔菌WN-3生长的最佳碳源为葡萄糖和甘露糖,最佳氮源为麦麸和玉米;菌株在pH6.5条件下生长最好,适宜生长温度为28℃。[结论]该方法研究了瓦尼木层孔菌WN-3分类地位的分子生物学验证及生理学特性,为进一步研究其人工栽培和规模化生产提供了理论基础。[ Objective ] To study molecular biological verification of taxonomic status and physiological characteristics of PheUinus vaninii WN-3. [ Method] The methods of molecular biology were used to verify the taxonomic status of strainWN-3. The physiological characteristics of WN- 3 were researched by growth rate method. [ Result] The results showed that: The length of rDNA ITS sequence of WN-3 is 756 bp. Sequence a-lignment in Genbank database showed the similarity of strain WN-3 and P. vaninii was more than 98%. So the strain WN-3 was verified as P. va-ninii. The Genbank accession number of WN-3 is HQ845057. Strain WN-3 can grow well in the PDA media which pH were 6.5, the optimum carbon sources for WN-3 growing were glucose and mannose and the optimum nitrogen sources were corn and wheat bran. The optimum tempera-ture for growing was 28 ℃. [ Conclusion ] Molecular biological verification of taxonomic status and physiological characteristics of Phellinus vani-nii WN-3 were studied, which will lay a theoretical basis for further study its artificial cultivation and large scale production.
