H2-Ab1基因在变应性鼻炎小鼠鼻黏膜中表达的研究

摘　　要：目的:研究H2-Ab1基因在变应性鼻炎小鼠鼻黏膜中的表达并与正常小鼠比较。方法:选取24只8周龄成熟雌性129/sv小鼠随机分成实验组、对照组,每组12只。实验组采用卵清蛋白(OVA)致敏激发,建立小鼠变应性鼻炎模型,对照组以磷酸盐缓冲液代替OVA。造模成功后取小鼠鼻腔黏膜固定后染色评估鼻黏膜嗜酸粒细胞、肥大细胞浸润等病理学变化,检测小鼠血清H2-AB1蛋白、IL-4和IFN-γ细胞因子水平和OVA特异性IgE抗体浓度。取鼻黏膜提取RNA行实时荧光定量PCR检测H2-Ab1mRNA的表达情况。结果:与对照组相比,实验组小鼠鼻腔黏膜纤毛部分脱落,黏膜下可见嗜酸粒细胞及肥大细胞浸润,血清中OVA特异性IgE、IL-4及H2-AB1浓度显著增高,IFN-γ浓度降低(P<0.05),H2-Ab1mRNA表达水平增高。结论:H2-Ab1mRNA及H2-AB1蛋白在变应性疾病中表达增高,提示HLA-DQB1基因对变应性鼻炎发病机制中的Th1/Th2失衡可能起到重要的调节作用。Objective.. To investigate the level of H2-Abl in the nasal mucosa of mice with allergic rhinitis. Method..Twenty-four female 129/sv mice were divided into 2 groups., ovalbumin（OVA） group and control. The allergic rhinitis models were induced by classical method with OVA. After the last challenge, the pathological differences between the two groups were surveyed. The levels of H2-AB1 were measured by ELISA and quantita- tive real time PCR. Result： The expression of H2-Ahl is higher in subjectswith AR than that in controls（P〈 0.05 ）. Conclusion： The levels of H2-Abl were highly increased in allergic rhinitis group, which might be associated with the pathogenesis of allergic rhinitiS.
