摘  要:目的观察小檗碱对高糖条件下血管内皮细胞迁移的影响,并研究可能的作用机制。方法以体外培养的EA.hy926人脐静脉血管内皮细胞为模型,采用划痕试验的方法研究高糖及不同浓度条件下对内皮细胞迁移的影响;采用电泳迁移率变动分析法检测核因子-κB(NF-κB)的DNA结合活性。结果 33mmol/L浓度的葡萄糖可抑制内皮细胞的迁移;小檗碱可逆转高糖对内皮细胞迁移的抑制作用。作用机制的研究表明,高糖可诱导NF-κB DNA结合活性上调,而采用小檗碱处理后可降低NF-κB活性。结论小檗碱可逆转高糖对内皮细胞迁移的抑制作用,该作用可能涉及抑制NF-κB的DNA结合活性上调。Objective To investigate the effect of berberine on the migration of endothelial cells treated by high glucose ,and explore the potential mechanisms . Methods EA .hy926 cell line was used in the present study . Cells were treated with 33 mmol/L D-glucose alone or combined with different con-centrations of berberine ;the migration rate of endothelial cells was measured by wound scratch assay . DNA binding activity of NF-κB was detected by using electrophoretic mobility shift assay (EMSA ) . Results 33 mmol/L D-glucose inhibited the migration of endothelial cells ,whereas berberine reversed the effect of high glucose on endothelial cells . Further mechanism studies showed that high glucose up-regu-lated the activity of NF-κB . Berberine inhibited the up-regulation of NF-κB activity induced by high glu-cose . Conclusion Berberine reverses the inhibition of D-glucose on endothelial cells ,w hich might be as-sociated with the suppression of up-regulation of NF-κB activity .
