摘 要:目的:研究不同负荷运动对慢性心力衰竭大鼠心肌细胞凋亡及相关蛋白表达的影响,探讨运动训练对心力衰竭后心肌的保护机制。方法:采用大鼠LAD结扎心肌梗死后心力衰竭模型,应用TUNEL、Western-blot法分别检测模型安静组(C+LAD组)、模型低强度运动训练组(LE+LAD组)、模型中等强度运动训练组(ME+LAD组)、模型高强度运动训练组(HE+LAD组)大鼠心肌细胞凋亡指数和心肌细胞凋亡相关蛋白Bax、Bal-2、Active-caspase-3、caspase-8表达。结果:与C+LAD组比较,LE+LAD组及ME+LAD组心肌细胞凋亡指数下降(P<0.05或P<0.01),HE+LAD组心肌细胞凋亡指数显著升高(P<0.05);LE+LAD组Active-caspase-3及caspase-8表达显著下降(P<0.05或P<0.01),Bcl-2表达显著升高(P<0.01);ME+LAD组Bax、Active-caspase-3和Caspase-8表达均显著下降(P<0.01),Bcl-2表达显著升高(P<0.01);HE+LAD组Bax表达显著升高(P<0.05)。结论:不同负荷运动训练对心力衰竭大鼠心肌细胞凋亡产生不同影响。适宜强度运动训练对心肌细胞的保护作用可能通过Bcl-2-Bax-Active-cas-pase3-Caspase8途径实现。Objective To investigate the impact of different intensity exercises on myocardial apoptosis and related protein expression in rats with chronic heart failure (CHF). Methods The CHF rat model was established by ligation of left anterior descending coronary artery (LAD)and resulted in myocardial infarction. The myocardial apoptosis rate and expression of apoptosis related proteins, including Bax,Bal-2,active-caspase-3 and caspase-8 were detected by TUNEL and Western-blot techniques in CHF model control group (C+LAD),CHF model plus low-intensity exercise group (LE+ LAD),CHF model plus moderate-intensity exercise group (ME+LAD),and CHF model plus high- intensity exercise group (HE+LAD). Results Compare with C+LAD group, myocardial apoptosis index decreased significantly in groups LE+LAD (P 〈 0.05)and ME+LAD (P 〈 0.01), and significantly increased in group HE+LAD (P 〈 0.05);active-caspase-3 and caspase-8 expressions significantly decreased and Bcl-2 expression significantly increased in group LE+LAD (P 〈 0.05, P 〈 0.01, and P 〈 0.01, respectively);and Bax expression significantly increased in group HE +LAD (P 〈 0.05). Conclusion Different intensity exercises affect myocardial apoptosis in chronic heart failure differently, and appropriate intensity exercise can protect the myocardium from apoptosis through Bal-2-Bax- active caspase3-caspase8 pathway.
