摘 要:目的:观察局部湿热对乳腺癌荷瘤小鼠肿瘤局部miRNA-7和miRNA-126表达的影响,并探讨其意义。方法:建立小鼠乳腺癌实验动物模型,进行局部湿热处理。28 d后,用Real-time PCR探针法分别检测肿瘤局部miRNA-7和miRNA-126的表达变化;同时用TUNEL法检测肿瘤细胞凋亡变化。结果:与对照组相比,局部湿热处理组中肿瘤组织miRNA-7和miRNA-126的表达均显著上调(P<0.05)。同时,肿瘤局部细胞凋亡率也明显增加(P<0.05)。结论:湿热治疗可改变肿瘤局部中特定miRNAs分子的表达,这可能是其导致肿瘤细胞凋亡的重要原因。Objective: To observe the effects of local wet hyperthermia on the expression of miRNA -7 and miRNA -126 in mouse mammary carcinoma and explore its significance. Methods: An experimental ani- mal model of mouse mammary carcinoma was established and then treated with local wet hyperthermia. On day 28, the relative expression of miRNA -7 and miRNA -126 was determined by real- time PCR assay. Moreo- ver, the apoptosis of cells in tumor mass was detected by TUNEL assay. Results: The expression of miRNA - 7 and miRNA - 126 in local wet hyperthermia treated group increased significantly compared with that in the control group (P 〈 0. 05). Moreover, the apoptosis rate of local tumor cells also increased significantly (P 〈 0. 05). Conclusion: Wet hyperthermia treatment could alter the expression of specific miRNAs molecules, which may be closely related to enhanced apoptosis of tumor cells.
