摘 要:目的:研究四逆散对海马神经元发生的影响。方法:采用皮质酮(CORT)抑制海马神经元发生的体外模型,在原代海马神经元培养体系中加入CORT后,运用MTT法测定神经元存活率,以5-溴脱氧尿嘧啶核苷(Brdu)标记增殖细胞,以神经元特异性烯醇化酶(NSE)标记成熟神经元生长。结果:四逆散含药血清能够提高CORT作用下海马神经元的存活率(P<0.05),增加了培养海马神经元Brdu数目(P<0.01)和NSE阳性细胞数量(P<0.05),并能促进突起生长。结论:四逆散含药血清可以有效地支持高皮质激素环境中神经元存活、增殖、生长,具有一定的促进海马神经元发生的作用。四逆散的抗抑郁作用与此机制有关。Objective: To study the effects of Sinisan on the hippocampal neurogenesis. Methods: The model of hippocampal neurogenesis inhibited by corticosterone was established in vitro. After adding corticosterone (CORT) into the culture system of the primary hippocampal neurons, MTT was used to analyze the survival rate of neurons. 5-bromodeoxyuridine (Brdu) labeling method was applied to identify the proliferating cells. Neuron-specific enolase (NSE) positive neurons were observed to evaluate the growth of neurons. Results: Serum containing Sinisan could improve the survival rate of hippocampal neurons cultured by excessive corticosterone (P〈0.05), increase the number of Brdu (P〈0.01) and NSE (P〈0.05) positive neurons, and promote the neurite outgrowth. Conclusion: Serum containing Sinisan could effectively enhance the survival, proliferation and growth of neurons in high corticosterone condition, and it has a certain effect of promoting hippocampal neurogenesis. The antidepressant effect of Sinisan is associated with this mechanism.
