寿胎丸对复发性流产模型小鼠子宫蜕膜HSP27和Tf表达的影响

摘　　要：目的:检测寿胎丸对复发性流产模型小鼠子宫蜕膜组织热休克蛋白27(HSP27)及转铁蛋白(Tf)表达的影响,探讨寿胎丸可能作用的环节。方法:建立复发性流产模型,用双相电泳和质谱技术等蛋白质组学技术,寻找出与复发性流产相关的差异蛋白质HSP27、Tf,用Western Blot和免疫组化实验检测寿胎丸对HSP27和Tf蛋白表达的影响。结果:Western Blot和免疫组化结果显示,小鼠模型组与正常组相比,蜕膜HSP27表达明显升高,Tf表达显著下降,差异有统计学意义(P<0.01);寿胎丸高、中、低剂量组与模型组相比,蜕膜HSP27表达均降低,差异有统计学意义(P<0.01),寿胎丸高剂量组与中、低剂量组相比差异有统计学意义(P<0.01)。寿胎丸高剂量组可升高复发性流产小鼠子宫蜕膜Tf的表达,差异有统计学意义(P<0.01);寿胎丸中、低剂量组子宫蜕膜Tf表达与模型组相比差异无统计学意义;寿胎丸高剂量组与中、低剂量组相比差异有统计学意义(P<0.01)。结论:寿胎丸通过调节上述蛋白的表达,从而实现维持妊娠,降低胚胎丢失率,这可能是其安胎的作用机制之一。Objective： To detect the effective mechanisms of Shoutaiwan on the expression of uterus decidual tissues HSP27 and Tf in recurrent spontaneous abortion model mice, and to explore to the possible role of the link. Methods： The preparation of recurrent abortion models, using two dimensional gel electrophoresis and mass specu＇ometry proteomics technology, to find out the differences in protein associated with recurrent spontaneous abortion of HSP27 and Tf. The expression effect of HSP27 and Tf with Sboutaiwan was detected by immunohistochemical and Western Blot analysis. Results： The expression of HSP27 in models group are higher than the normal pregnancy group, and Tf in models group are lower than the normal pregnancy group, the difference is statistically significant （P〈0.01）, the expressions of HSP27 in Shoutaiwan low-dose group, middle-dose group and high-dose group are lower than the model group, the difference was statistically significant （P〈0.01）. Shoutaiwan high-dose group compared with Shoutaiwan low-dose group, middle-dose group, which was highly statistically significant （P〈0.01）. Shoutaiwan high-dose group increased the expression of recurrent spontaneous abortion mouse uterine decidual Tf, the difference was highly statistically significant （P〈0.01）. Decidua Tf expressions of Shoutaiwan low-dose group and middle-dose group were no significant difference compared with model group. Conclusion： Shoutaiwan, through the protein expression, achieves the maintenance of pregnancy, reducing the rate of embryo resorption, which may be one of the mechanisms of preventing miscarriage effect.
