摘 要:目的:观察前列宁胶囊(QC)对良性前列腺增生(BPH)大鼠血清雄激素、细胞外基质(ECM)含量及前列腺组织中雄激素受体(AR)、ECM表达的影响,探讨性激素、ECM在BPH中的影响及QC治疗BPH的机制。方法:SD大鼠随机分为空白组,BPH模型组,QC低、高治疗组(2.25、9g/kg)。建立大鼠BPH模型,给予QC干预。28d后ELISA法检测大鼠血清睾酮(T)、纤维黏连蛋白(FN)、胶原蛋白Ⅳ(CollagenⅣ),RT-PCR法、免疫组化法检测大鼠前列腺组织中AR、FN、CollagenⅣ的mRNA、蛋白表达。结果:QC各剂量组能明显降低血清中的FN、CollagenⅣ含量,提高雄激素含量(P<0.05,P<0.01),抑制前列腺组织AR、FN、CollagenⅣmRNA与蛋白表达,与模型组比较有显著性差异(P<0.01)。结论:QC对BPH大鼠有明显的治疗作用,能抑制前列腺组织中AR、FN、CollagenⅣ表达,表明调节AR、FN、CollagenⅣ表达可能是其治疗BPH的重要机制。Objective: To observe the influence of Qianliening Capsule (QC) on the levels of serum testosterone and extracellular matrix (ECM) in rats with benign prostatic hyperplasia (BPH), and the expression of androgen receptor (AR) and ECM in prostatic tissue, as well as to investigate the effects of QC on testosterone, AR, ECM and the possible molecular mechanisms mediating its anti-BPH activity. Methods: Fourty male Sprague-Dawley (SD) rats were randomly divided into four groups: control group, BPH model group, QC low dose group (2.25g/kg), QC high dose group (9g/kg). The rats were generated to BPH, and intervened with QC. After 28 days, testoids (T), fibronectin (FN), collagen Ⅳ in serum were examined by ELISA. The mRNA expression of AR, Collagen IV and FN in prostatic tissue were determined by RT-PCR, and the protein expressions were determined by immunohistochemistry methods. Compared with model group, serum FN, Collagen Ⅳ were significantly reduced and serum T in QC-treated groups were elevated significantly (P〈0.05, P〈0.01); Compared with the model group, the mRNA and protein expression of AR, FN and Collagen Ⅳ in QC-treated groups were decreased significantly (P〈0.05). Conclusion: QC has a significant therapeutic effect on BPH rats. QC can inhibit the expression of AR, FN and Collagen IV in prostate tissue. It indicates that regulation of AR, FN and Collagen Ⅳ may be one of the mechanisms of QC in treating BPH.
