摘  要:目的:探讨芪参颗粒联合运动疗法对慢性心力衰竭大鼠心肌细胞凋亡的影响及其作用机制。方法:采用大鼠左冠状动脉结扎心肌梗死后心力衰竭模型,应用TUNEL、Western-blot法分别检测模型组和芪参颗粒组、芪参颗粒联合运动疗法组、单纯运动疗法组大鼠心肌细胞凋亡率和心肌细胞凋亡相关蛋白Bax、Bcl-2、activecaspase-3、caspase-8表达情况。结果:芪参颗粒组和芪参颗粒联合运动疗法组能够显著降低心力衰竭大鼠梗死边缘区心肌细胞凋亡率(P<0.01),其机制可能与上调Bcl-2水平(P<0.01),抑制Bax、active-caspase-3、caspase-8水平(P<0.01)相关,而单纯运动组可能通过上调Bax、active-caspase-3导致上述区域心肌细胞凋亡率增加。结论:芪参颗粒联合运动疗法能够显著抑制心力衰竭大鼠心肌细胞凋亡。Objective: To investigate the myocardial protection mechanism of Qishen Granules combined with exercise therapy on apoptosis and related protein expression in heart failure rats. Methods: Rat LAD ligation model of heart failure after myocardial infarction were used, and TUNEL, Western-blot methods were used to detect the myocardial apoptosis rate and apoptosis related proteins Bax, Bcl-2, Active-caspase-3, caspase-8 expression in the model control group, Qishen Granules group, Qishen Granules combined with exercise therapy group, exercise therapy group. Results: Qishen Granules and Qishen Granules combined with exercise therapy could significantly reduce apoptosis rate (P〈0.01) in heart failure rats, which may be related to the increase of the level of Bcl-2 (P〈0.01), and the inhibition of Bax, active-caspase-3, caspase-8 (P〈0.01). The exercise training mightlead to the region increased apoptosis rate by the up-regulation of Bax and active-caspase-3 level. Conclusion: Qishen Granules combine with exercise therapy could inhibit the myocardial apoptosis in heart failure rats.
