摘  要:目的:探讨大鼠胰组织中是否表达卵泡刺激素(FSHl3)及其受体(FSHR),明确胰细胞内是否存在卵泡刺激素自分泌调节现象和具有合成内源性卵泡刺激素的能力。方法:采用RT-PCR法探讨大鼠胰是否存在FSH及其受体mRNA,原位杂交方法对FSH及FSHR进行细胞定位,并运用其克隆获得的基因cDNA的核心序列进行分析。结果:大鼠胰岛大部分细胞及部分胰外分泌部细胞含有FSH和FSHRmRNA杂交信号,阳性物质分布于细胞质,细胞核呈阴性反应。结论:胰细胞自身具有合成和分泌内源性卵泡刺激素的功能,存在卵泡刺激素自分泌调节现象,说明FSH对胰细胞的功能作用可能通过旁分泌或自分泌的作用实现。Objective: To detect the expressions and localization of follicle stimulating hormone (FSH) β and its receptor in rat pancreas, in order to evaluate whether pancreas cells of rats have capability of autocrine regulation and synthesis of endogenous FSH. Methods: RT-PCR was used to detect the mRNA expression of FSH and its receptor, and in situ hybridization was used to localize these mRNA in rat pancreas. Then, the core sequences of FSH and its receptor were cloned and analyzed. Results: The mRNA of FSH and its receptor was detected in most cells of islet and partial pancreas exocrine cells. And, these mRNA was dominantly localized in cytoplasm, not in nucleus. Conclusion: These findings suggest that the cells in rat pancreas could have capability to synthesize and secrete the endogenous FSH and its receptor. And, FSH may be involved in functional regulation of pancreas through autocrine and paracrine approaches.
