摘 要:目的 在重组人肿瘤坏死因子-α刺激基因-6干扰前后,检测病理性瘢痕成纤维细胞内Bcl-2、Bax的表达情况及细胞凋亡情况,探讨肿瘤坏死因子-α刺激基因-6与病理性瘢痕形成的关系及其机制.方法 增生性瘢痕和瘢痕疙瘩组织各5例,以组织块贴壁法培养成纤维细胞,待细胞培养传代至第3、4代后,以TUNEL法和透射电镜法检测重组人肿瘤坏死因子-α刺激基因-6干扰对成纤维细胞凋亡的影响,以免疫组化法检测各组成纤维细胞内Bcl-2和Bax表达的变化.结果 TUNEL和透射电镜显示,重组人肿瘤坏死因子-α刺激基因-6促进病理性瘢痕成纤维细胞的凋亡.免疫组化显示,病理性瘢痕成纤维细胞内的Bcl-2表达减少,而Bax表达增多.结论 肿瘤坏死因子-α可抑制病理性瘢痕成纤维细胞的增殖,并促进其凋亡,其机制可能与其调节成纤维细胞内Bcl-2和Bax的表达有关.Objective To examine the effect of tumor factor alpha stimulated gene-6 on the Bcl-2, Bax ex- pression and apoptosis in the fibrohlasts of pathological scar and explore relationship between the tumor factor alpha stimulated gene-6 and formation of pathological sear and its formational mechanism. Methods The samples were harvested from hyperplastic scar and keloid, and then were cultured to fibroblasts with adherence method. The fibro- blasts were treated with tumor factor alpha stimulated gene-6 at 3rd and 4th passage, and then the fibroblasts apopto- sis were detected by TUNEL and transmission electron microscope methods and the expression of Bcl-2 and Bax was detected by immunohistochemisty in the two groups. Results Tumor factor alpha stimulated gene-6 significantly pro- moted cell apoptosis of pathological sear fibroblasts by TUNEL and transmission electron microscope. The expression of Bcl-2 in the fibroblasts of pathological scar decreased, while the expression of Bax increased by IHC. Conclu sion The pathological scar formation is associated closely with tumor factor alpha stimulated gene-6. Tumor factor alpha stimulated gene-6 may inhibit the formation of pathological scar by iIflaibiting pathological scar fibroblasts' pro- liferation and promoting its apoptosis, which may he related with the expression of Bax and Bel-2.
