摘 要:LEAFY(LFY)基因是花分生组织形成的必需基因,在成花过程中发挥重要作用。本研究以荷花品种‘红霞满天’为材料,利用已经获得的荷花LFY基因片段,采用RT-PCR和RACE技术克隆得到荷花LFY基因cDNA序列,命名为NnLFY。NnLFY全长cDNA为1 494 bp,开放阅读框(ORF)为1 173 bp,编码390个氨基酸,预测其相对分子量为44 517.1 Da。与其他物种已知的LFY基因序列进行同源性对比分析并构建系统进化树,结果显示荷花LFY基因与甜瓜LFY基因的亲缘关系最近。实时荧光定量PCR结果表明,LFY基因在荷花幼叶期、花蕾期、盛花期的根、茎、叶、花中均有表达,其中,花蕾期各器官的表达量较高。LEAFY(LFY) genes are a class of inflorescence meristem identity gene which play important roles in the process of flowering.In this study,a homologue of LFY,designated NnLFY,was isolated from Nelumbo nucifera ‘Hongxiamantian’ by reverse transcriptase-polymerase chain reaction(RT-PCR) and rapid amplification of cDNA ends(RACE).The cDNA sequence of NnLFY was 1494 bp long and contained an open reading frame(ORF) of 1173 bp encoding a protein of 390 amino acids.Its molecular mass was 44 517.1 Da.The NnLFY shared highest homology with the LFY from melon according to the homology and phylogenetic analyses of the relationship of NnLFY with some known LFY genes from other plants.Real-time quantitative PCR analysis indicated that the NnLFY was ubiquitously expressed in the root,stem,leaf and flower in the different periods and the highest expression was detected in the bud period.
