摘  要:为监测猪繁殖与呼吸综合征病毒(PRRSV)的遗传变异及流行情况,本研究采用猪肺泡巨噬细胞培养分离PRRSV野毒株,从某疑似猪繁殖与呼吸综合征发病猪场的发病猪肺脏病料中分离了一株PRRSV,命名为HEL-12。并将该病毒测序分析,其基因组序列已提交到NCBI (序列号:KJ019330)。基因序列比对显示,该分离株与经典株相比其NSP2蛋白存在HP-PRRSV 30个氨基酸缺失的典型特征,与HP-PRRSV代表株JXA1和HuN4株的同源性仅为97.3 %,其变异主要位于基因组编码的结构蛋白GP2~GP5和非结构蛋白NSP2。血清中和试验表明10份HP-PRRSV疫苗抗血清均不能中和该病毒株,表明HEL-12的抗原发生了较大变异,是一株新的HP-PRRSV变异株。本研究表明,PRRSV一直处于不断变异中,需要对其遗传变异持续监测。To investigate the mutations of porcine reproductive and respiratory syndrome virus (PRRSV), a PRRSV (designated HEL-12) was isolated with porcine alveolar macrophage cells from the lung tissue of a affected swine herd and subject to whole genome sequencing. The alignment result showed that the virus isolate had the typical molecular feature of high pathogenic PRRSV(HP-PRRSV), characterized with a 30 amino acid encoding sequence deleted on the NSP2 gene. However, comparing to reference HP-PRRSVs in GenBank, the isolate of HEL-12 was only 97.3% nucleotide identity with HP-PRRSV strains of JXA1 and HuN4, of which a lot of point mutations were found mainly with in encoding regions of the NSP2 and GP2-GP5. Furthermore, the virus neutralization test revealed that the virus isolate was unable to be neutralized with the serum derived from HP-PRRSV vaccine(HuN4-F112 strain) immunized piglets, which indicated the crucial antigens for inducing neutralization antibody had mutated in the isolate of HEL-12, suggesting the virus isolate is a new variant HP-PRRSV.
