摘 要:为研究鸭源致病性大肠杆菌(E.coli)O46分离株感染雏鸭后在其体内组织器官中的动态分布,本实验将E.coli O46分离株经口服、肌注和皮下注射3种途经感染雏鸭,于感染后不同时间点采集雏鸭15个不同的组织器官制备组织切片,采用免疫组化染色方法,利用E.coli O46定型血清对样品中细菌抗原进行定位跟踪检测。结果显示:口服、肌注和皮下注射3种途经接种E.coli O46分离株后,15个不同的组织器官中除气管未检测到细菌抗原外,其余的组织器官均检测到细菌抗原。心脏、肺脏、脾脏、肾脏和肠道是感染的主要靶器官,并且抗原主要存在于其感染细胞的细胞质中,阳性信号最早出现于心脏。本研究表明鸭源E.coli O46分离株能够在鸭体内广泛的侵嗜增殖并造成鸭急性发病和死亡。To investigate the dynamic distribution of a pathogenic duck E.coli O46 strain in different organs of ducklings after experimental infection, the ducklings were inoculated with the E.coli O46 isolate via oral, intramuscular injection and subcutaneous routes, respectively. At different time points after infection, tissue samples were collected from 12 organs, and slices were prepared and tested by immunohistochemical assay with O46-specific antiserum to observe the bacterial antigen localizations in infected ducklings. The results showed that the bacterial antigen was detected in all organs except for trachea. Heart, lung, spleen kidney and intestinal tract were proven to be the main target tissues of the bacteria, and bacterial antigen mainly located in cytoplasm of infected cells. Positive signals were first detected in heart. The results obtained in this study suggested that the E.coli O46 strain isolated from duck has a broad tissue tropism, and therefore could lead to acute onset and death of the ducks.
