摘 要:目的:利用同源克隆从海岛棉中克隆了1个新的乙烯应答因子(ethylene-responsive factors ERF)为研究该基因功能奠定基础。方法:利用RNA提取试剂盒提取海岛棉叶片总RNA,采用RACE(rapid amplification of cDNA ends,RACE)技术获得基因全长,利用分子生物学软件进行序列分析,利用q-PCR分析基因在乙烯诱导下的表达情况。结果:cDNA全长序列为926bp(GenBank登录号为:KF555288),开放阅读框为624bp,编码208个氨基酸,命名为GbERFa。乙烯诱导在6h表达量最高。结论:该基因参与乙烯信号代谢途径,在棉花防卫反应中可能起一定的作用。Objective: A novel ERF gene ( ethylene - responsive factors ERF) was cloned from C, ossypium barbadense. Leaves. That is the foundation for the further function study. Method: The total RNA was extracted by RNA Extraction Kit. The full - length gene sequence was obtained using homology - based cloning by RACE ( rapid amplification of cDNA ends, RACE) technology, the sequence was analysised with the molecular biology software, and gene expression was analysised under the ethylene induced using q - RT PCR. Result: The full length cDNA sequence is 926bp( GenBank Accession was No. JQ659260). The open reading frame ( ORF ) is 624bp encoding a residue of 208 amino acids, named GbERFa. The gene expression was highest at 6h under the ethylene induced. Conclusion: The gene involved in ethylene signaling pathways, and may play a certain role in cotton defense reactions.
