摘 要:噬菌粒载体是包含质粒复制起点的丝状噬菌体衍生载体。噬菌粒通常只编码一种丝状噬菌体的外壳蛋白,其他完成生命周期必须的结构和功能蛋白由辅助噬菌体提供。噬菌粒是噬菌体展示技术最常用的表达载体,它的诸多优点决定了噬菌粒在噬菌体展示领域中具有广阔的应用前景。文章概述了噬菌粒载体的结构、生命周期和主要元件,着重介绍了载体的优化策略及应用,最后对其应用前景进行了展望,以期为进一步优化和利用噬菌粒载体提供参考。Phagemid vectors are filamentous - phage - derived vectors containing the replication origin of a plasmid. Phagemid vectors often encode only one kind of coat protein, other structural and functional proteins necessary to accomplish the life cycle are provided by the helper phage. Phagemid vectors were the most common vectors used in phage display, it had many merits decision phagemid vector had broad application prospects in the field of phage display. In this review, we summaried phagemid vector structure,life cycle in phage display and characteristics of phagemid vector. We mainly emphasized construction strategies and application. Finally, we made some prospect for the further application. We expect that our experience can provide references for optimizing and using of phagemid vector perfectly.
