摘 要:目的探讨铅暴露对不同发育阶段仔鼠海马中谷氨酰胺合成酶(GS)活力及蛋白表达的影响。方法将24只孕鼠随机分为4组,分别为对照(蒸馏水)组及0.5、1.0、2.0 g/L铅暴露组,每组6只。采用自由饮水方式进行染毒,母鼠从妊娠第1天至仔鼠断乳(出生后21 d左右);仔鼠从断乳后至出生后(PND)40 d。分别在PND10、20和40 d,测定仔鼠脑组织中铅含量、海马组织中GS活力及其蛋白表达。结果随着铅暴露浓度的升高,仔鼠脑组织中的铅含量呈上升趋势,海马组织中GS活力及其蛋白表达呈下降趋势。结论饮水铅暴露可通过抑制不同发育阶段仔鼠海马中GS蛋白的表达,导致海马中GS活力下降。Objective To investigate the effects of lead exposure on glutamine synthase (GS) activity and protein expression in the hippoeampus at different developmental stages. Methods Twenty-four female pregnant mice were randomly divided into four groups: control (distilled water)group,0.5,1.0 and 2.0 g/L lead-exposed group,six in each. The pregnant mice were given lead acetate through drinking water from the first day of gestation to weaning (about postnatal day 21) and the pups from weaning to postnatal day 40 drank the same water. Lead content in the brain,GS activity and protein expression in the hippocampus were measured in postnatal day (PND)IO,20 and 40. Results With the increase of lead exposure,the lead content in the brain increased, GS activity and protein expression in the hippocampus decreased. Conclusion Lead exposure through drinking water may inhibit the expression of GS protein in hippocampus in different develoomental stazes of mice.
