摘 要:目的探讨化瘀理肺方对博莱霉素所致大鼠肺纤维化的防治作用与机制。方法雄性Wistar大鼠84只,随机分为模型组、中药组、激素组与对照组,每组21只。前3组用1%戊巴比妥钠(35 mg.kg-1)腹腔注射麻醉后,气管插管注入博莱霉素(5 mg.kg-1)建立纤维化模型,对照组气管内注入0.25 ml生理盐水,次日开始中药组给予化瘀理肺方17 190 mg.kg-1(2 ml)灌胃,模型组与对照组给予生理盐水2 ml/只灌胃,激素组腹腔注射氢化可的松琥珀酸钠(25mg.kg-1),于d 7、d 14和d 28各组随机处死7只。取肺组织行HE染色进行病理学观察,检测各组肺组织均浆中GSH、SOD及MDA含量。结果化瘀理肺方能明显减轻肺部炎症及纤维化,并能提高肺组织均浆中GSH、SOD的含量,降低MDA的含量。结论化瘀理肺方可通过抗氧化作用减轻博莱霉素致肺部炎症并抑制肺纤维化的发展。Objective The objective of the study was to explore the preventive effect and mechanism of Huayulifei on bleomycin-in- duced pulmonary fibrosis in rats. Methods 84 male Wistar rats were randomly divided into a model group, a traditional Chinese medicine group, a hydrocortisone group, and a control group; each group was consisted of 21 rats. The former three groups were anesthetized by abdominal injection with 1% pentobarbital sodium (35mg.kg-l), followed by intratracheal administration with bleomycin (5mg.kg-1) to create a pulmonary fibrosis model; the control group was intratracheally administered with 0.25 ml of saline instead. On the following day, the traditional Chinese medicine group were orally garaged with 17190mg.kg-1 of Huayulifei (2ml), the model group and the control group were gavaged with 2 ml of saline, and the hydrocortisone group was intraperitoneally injected with hydrocortisone sodium suceinate (25mg.kg-l). 7 rats from each group were randomly chosen and sacrificed on the 7~, 14~, and 28~ days. Lung tissues of the rats were stained with hematoxylin and eosin (HE) for pathological analysis, and the contents of GSH, SOD, and MDA in the homogenate of the lung tissues were determined. Results Huayulifei significantly ameliorated lung inflammation and pulmonary fibrosis, increased the contents of GSH and SOD, and decreased MDA level in lung tissue homogenates. Conclusion Huayulifei may exert antioxidative effects that reduce bleomycine-indueed lung inflammation and suppress the progression of pul- monary fibrosis.
