摘 要:目的 探讨增龄对大鼠骨骼肌细胞葡萄糖转运蛋白 4(glucosetransporter 4,GLUT4 )的影响。 方法 SD实验大鼠分为 2组 :青年组 (3月龄 )和老年组 (2 4月龄 )各 10只。制备GLUT4 羧基端12肽的多克隆抗体 ,利用Western印迹法检测 2组大鼠骨骼肌细胞GLUT4 蛋白含量 ,并检测大鼠尾静脉血糖。 结果 老年组大鼠的血糖 (5 6± 0 5mmol/L)略高于青年组大鼠 (4 5± 0 5mmol/L) ,但差异无显著性 ;青年组大鼠骨骼肌细胞内GLUT4 相对含量为 10 9 6 2± 12 2 5 ,而老年组为 86 46± 8 2 5 ,差异有显著性 (P <0 0 5 )。 结论 增龄可引起大鼠骨骼肌细胞GLUT4 蛋白含量明显减少 ,使骨骼肌细胞对葡萄糖的转运发生障碍 ,这可能是老年人易产生胰岛素抵抗的机制之一。Objective To investigate the difference of glucose transporter 4 (GLUT 4) protein content in skeletal muscles of young and aged rats. Methods Twenty SD rats were divided into two groups, namely young rats (3 months) and aged rats (24 months). The content of GLUT 4 protein in skeletal muscle cells was detected by Western blot analysis in two group rats. The polyclonal antibody against GLUT 4 protein was obtained from GLUT 4 protein immunized rabbits. Blood glucose level of two groups was also detected. Results The relative content of GLUT 4 protein in skeletal muscle cells of aged rats (86 46±8 25) decreased 21 1% as compared with young one (109 62±12 25) ( P <0 05). There was no significant difference in blood glucose level between two groups. Conclusions The decreased content of GLUT 4 protein in skeletal muscles cells may associated with age related insulin resistance in aged.
