摘 要:本研究采用DNA池直接测序法寻找京海黄鸡BMP-4基因外显子4区域的多态位点,用PCR-SSCP技术检测发现的多态位点在京海黄鸡中的单核苷酸多态性,并分析其对京海黄鸡生长和繁殖性状的遗传效应。结果表明:在外显子4区域检测到2个突变位点(C701T,A967G);C701T位点经PCR-SSCP分析显示2种基因型(AA和AB),其中AA型为优势基因型。最小二乘分析表明,C701T位点对京海黄鸡的生长和繁殖性状均无显著影响(P>0.05)。The mutations in the exon 4 of the BMP-4 gene were detected in Jinghai Chicken using DNA pooling and sequencing. Single nucleotide polymorphisms (SNPs)of the found mutations were detected in Jinghai Chicken by PCR- SSCP. The genetic effects on growth and reproduction traits were also analysed. Results showed that two SNPs (C701T, A967G) were detected in the exon 4 of the BMP-4 gene. Two genetypes (AA and AB) were displayed by PCR-SSCP, and the genotype AA was the predominant genotype. The least square means analysis showed the differences for growth and reproductive traits among the 2 genotypes in Jinghai chicken were nonsignificant (P〉0.05).
