摘  要:本研究采用PCR-RFLP方法对梅山猪Mx1基因第14外显子Hin6 I酶切位点多态性进行分析,同时对机体部分重要细胞因子(IFN-α、IFN-γ、IL-2、IL-6、IL-10和IL-12)水平以及早期生长性能(初生重、20日龄重以及40日龄重)进行测定,进一步分析梅山猪Mx1基因第14外显子多态性与细胞因子水平和早期生长性能间的关系,探讨将该酶切位点标记运用于梅山猪抗病育种实践的可行性。结果表明:梅山猪Mx1基因第14外显子Hin6 I酶切位点存在G36T突变,共检测到3种基因型,分别定义为AA、AB和BB。关联分析结果表明,AA基因型个体的IFN-γ水平显著高于AB基因型个体(P<0.05),也高于BB基因型的个体(P>0.05);3种基因型个体间其他细胞因子水平以及对应的早期生长性能间差异不显著(P>0.05)。结果提示,针对梅山猪群体Mx1基因Hin6 I酶切位点的分子选育可能会在一定程度提高机体的抗病毒能力,而不会对机体一般抗病力和生长发育产生不利影响。This study was conducted to analyze polymorphisms in exon 14 of Mxl gene by PCR-RFLP, determine level of the partial vital cytokines (IFN-α, IFN-γ, IL-2, IL-6, IL-10, IL-12) and early growth performance (birth weight, 20d weight and 40d weight), further analyze the association of polymorphisms in exon 14 of Mxl gene with significant cytokines level and early growth performance, discussing feasibility of Hin6I restriction site as genetic markers applied to breeding for disease resistance in Meishan pigs. The results showed that there were AA, AB and BB genotypes in Meishan pigs, while a G36T mutation was found in BB genotype. The correlation analysis indicated that IFN-γ level of AA genotype was significantly higher than that of AB genotype (P〈0.05), which supernal compared with that of BB genotype (P〉0.05); while other cytokines level and early growth performance had no significant difference in three kinds of genotypes between individuals (P〉0.05). The results pointed out that molecular breeding based on the point could not make unfavorable influence on resistance to disease and growing development, but may enhance anti-virus capability in Meishan pigs.
