摘 要:为探讨复方中药合剂对蛋仔鸡免疫功能的影响,选用7日龄健康海兰褐蛋仔鸡480只,随机分为4组,每组5个重复,复方中药合剂按0.5%、1.0%、1.5%添加饮水中,7日龄接种新城疫Clone30活疫苗,试验期为4周。分别于14、21、28日龄和35日龄,每组随机抽取10只鸡,测定胸腺、法氏囊和脾脏指数和新城疫抗体效价。35日龄采用MTT法测定淋巴细胞增殖率,脾脏、法氏囊石蜡组织切片,H·E染色,显微观察、测量和拍照。结果表明:复方中药合剂能显著或极显著提高蛋仔鸡胸腺、法氏囊和脾脏指数(P<0.05 or P<0.01);显著或极显著地提高T淋巴细胞增殖率(P<0.05 or P<0.01);显著提高血清抗体效价(P<0.05);显著或极显著提高脾小体平均直径、动脉周围淋巴鞘平均面积和法氏囊淋巴滤泡平均面积(P<0.05 or P<0.01);试验组与对照组比较,脾小体数量增多,脾动脉周围淋巴鞘明显增厚,鞘内淋巴细胞密集;法氏囊内淋巴滤泡数量较多,排列紧密,法氏囊皱襞发达,皱襞内淋巴细胞增多。结果表明,复方中药合剂能显著地促进蛋仔鸡免疫器官发育、淋巴细胞的增殖和疫苗抗体的产生,增强机体的免疫功能。In order to study the effects of preparation of traditional Chinese herbal medicine on immune function in egg-laying chickens. 480 healthy egg-laying chickens at the age of 7 days old were randomly divided into 4 groups with 5 repeates. The control group was watered normally, and the experimental groups were watered with 0.5%, 1.0% and 1.5% the preparation of traditional Chinese herbal medicine respectively. ND vaccine was immunized in every group at 7 days old. On 14, 21, 28 and 35 days old after gavaging, 10 egg-laying chickens were sampled randomly from each group. Then immune organs index of thymus, fabrieius and spleens, ND antibody titer were measured. At the age of 35 days, MTT method was used to determine the lymphocyte proliferation rate. fabricius and spleens werefixed. Paraffin sections were prepared, stained with HE, observed and determined under microscope and taken photographs. The results showed that the preparation of traditional Chinese herbal medicine could significantly improve the immune organ indexes(P〈0.05 or P〈0.01, the lymphocyte proliferation rate(P〈0.05 or P〈0.01), ND antibody titer (P〈0.05), the diameters of spleen node , the area of periaterial lymphatic sheath and the area of lymphoid follicle of fabricius(P〈0.05 or P〈0.01). The egg-laying chickens in experiment groups had following changes compared with the control group. The structure of spleen was amount of splenic nodule increased in white pul. Splenic periarterial lymphatic sheath thickening, lymphocyte becomes more and more intensive. The amount of lymphoid nodule in bursa of fabricius increased and arranged closely. Folds of fabricius has more developed and increased in Lymphocyte numbers. These results suggested that the preparation of traditional Chinese herbal medicine could stimulate development of immune organ, T lymphocyte proliferation and improve antibody titer, enhance the immune function in egg-laying chickens.
