壮精合剂对围绝经期综合征初老大鼠性激素的影响

摘　　要：目的:观察壮精合剂不同剂量对自然衰老型围绝经期综合征初老大鼠性激素及子宫、卵巢组织形态的影响。方法:40只13月龄雌性初老大鼠,随机分为空白对照组、壮精合剂高剂量组、壮精合剂中剂量组、壮精合剂低剂量组,每组10只。另选10只5月龄雌性大鼠作为青年对照组。壮精合剂各剂量组分别灌胃相应的药物,每日1次,连续4周,并于末次给药24 h后对大鼠进行取血后处死,分离子宫、卵巢。检测各组血清雌激素(E2)、卵泡刺激素(FSH)、黄体生成素(LH)、催乳素(PRL)水平,光镜观察卵巢、子宫形态改变。结果:与空白对照组比较,壮精合剂高、中剂量组能增加围绝经期综合征初老大鼠血清E2水平(P<0.05),壮精合剂各剂量组均能降低血清FSH、LH水平(P<0.01),增加PRL含量(P<0.01);与青年对照组比较,壮精合剂低、中剂量组E2水平明显降低,具有显著性差异(P<0.01),其余指标无统计学意义。壮精合剂各剂量组大鼠卵巢内卵泡发育状况有所改善,子宫内膜有一定的增厚,尤以壮精合剂高剂量组作用较为明显。结论:壮精合剂能有效改善围绝经期综合征初老大鼠的症状,其作用可能与调节体内性激素有关。Objective： To observe the effects of different doses of Zhuang Jing Mixture （ZJM） on natural aging old rats with early peri-menopausal syndrome. Method： Forty 13-month-old control group,high-mid-low dose group of ZJM,with primary senile female rats were randomized to 4 groups, blank 10 rats in each group. Another ten 5-month-old female rats were selected as young control group. After each group were infused with corresponding drugs by gavage once daily for 4 weeds continuously, the primary senile female rats were executed after the final medication and the blood were sampled. The uterus,ovaries were separated,and the levels of serum estrogen （E2）,follicle stimulating hormone （FSH）, luteinizing hormone （LH）, prolactin （PRL） of each group were detected,the morphological change of the uterus and ovaries were observed by light microscope. Results： Compared with blank control group,high-and mid- dose group of ZJM can increase the level of serum E2 of primary senile rats with perimenopausal syndrome（P〈0.05）, and all dose group of ZJM can decrease the serum FSH and LH levels（P〈0.01 ） and increase the level of serum PRL （P〈0.01）. Compared with young control group,the serum E2 level of low- and mid- dose group were significantly decreased （P〈0.01）,other index had no statistic significance. All dose group of ZJM can improve the condition of follicle development in the ovary, and thicken the endometrium to a certain degree. The effect of high dose group of ZJM was more obvious. Conclusion： ZJM can effectively improve the symptoms perimenopausal senile female rats,which is probably linked with sexual hormones regulation.
