摘  要:目的探讨在海水淹溺性肺损伤中肺组织STE20相关脯氨酸丙氨酸蛋白激酶(SPAK)表达的变化及在肺损伤发生发展中的作用。方法健康雄性SD大鼠30只,分为5组:对照组,海水处理1 h组,海水处理3 h组,海水处理6 h组,海水处理12 h组,每组6只动物。经气管缓慢注入4 ml/kg海水。处死后检测肺湿干比(W/D)、肿瘤坏死因子-α(TNF-α)和干扰素-1β(IL-1β)、SPAK mRNA及蛋白的表达。结果经气管海水灌注后,肺的湿干比较对照组显著升高。TNF-α和IL-1β的水平明显增高。RT-PCR及western blot方法检测肺组织匀浆中的SPAK转录及翻译均有上调。肺的湿干比,海水处理6 h组与对照组比较差异具有统计学意义(P<0.05);海水处理1 h,3 h组与对照组比较差异具有统计学意义(P<0.001 vs.P<0.01)。结论在海水高渗透压的刺激下,细胞通过转录和翻译上调SPAK的表达,参与肺的炎症反应,加重了急性肺损伤的程度。但是其确切的作用有待进一步的研究证实。Objective To observe the expression of STE20/SPSl-related proline/alanine-rich kinase (SPAK) in seawater inhalation induced acute lung injury and explore its effect on the occurrence and progression of acute lung injury. Methods A total of 30 Sprague-Dawley ( SD ) rats were randomly divided into 5 groups: control group, 1 h, 3 h, 6 h and 12 h seawater challenged groups(n =6). After challenged by endotracheal instillation of seawater (4 ml/kg) to establish lung injury models, the lung tissues wet-to-dry ratio, tuinor necrosis factor-α(TNF-α) , interferon-1 β( IL-1 β) , expression of SPAK were measured. Results The lung tissues wet-to-dry ratio was elevated significantly after seawater instillation, the same as TNF-α and IL-1β in the lung tissues. And the transcription and translation of SPAK were up-regulated detected by RT-PCR and western-blot. The lung tissues W/D ratio of the 6 h seawater challenged group had significant differences compared with control group ( P 〈 0.05 ) , and the same as the 1 h, 3 h group compared with control group ( P 〈 0. 001 vs. P 〈0.01 ). Conclusions The lung cells can participate in inflammatory response by up-regulating the transcription and translation of SPAK when challenged with seawater, and this reaction can aggravate the development of acute lung injury. However, this effect needs to be confirmed by further studies
