摘 要:目的利用盐固体分离培养基,从西藏自治区澜沧江边康宁镇一个47℃的盐井样品中分离纯化到一株耐热嗜盐菌菌株YJ0232。方法通过形态观察、生理生化特性和16S rRNA基因序列分析,鉴定嗜盐菌菌株YJ0232分类学地位。结果菌株YJ0232初步鉴定为中度嗜盐菌,属于盐单胞菌属(Halomonas sp.)菌株。其16S rRNA基因序列已被GenBank数据库收录,序列号为EU029645。结论本研究对澜沧江高盐环境微生物资源进行了初步探索研究。可为今后研究同类极端环境中新的物种资源以及微生物多样性提供参考。Objective To isolate a strain of thermotolerant halophilic bacteria YJ0232 from a 47 ℃salty well at Kangning, a town located at the side of Lancang River in Tibet Autonomous Region by using salt solid isolation me- dium. Methods Based on its physiological and biochemical properties, homology and phylogenetic analysis of 16S rRNA sequence, the taxonomic position of the halophilic strain YJ0232 was identified. Results The strain YJ0232 was identified as a moderate halophile bacterium strain and belonged to the genus of Halomonas. The GenBank ac- cession number of the 16S rRNA sequence of YJ0232 was EU029645. Conclusion The research is a preliminary exploration on the microbiological resources in high salt environment of Lancang River. The results will provide ref- erence information for further studies in this area.
