摘 要:目的:研究NSF对5/6肾切除大鼠模型肾纤维化干预作用及机制。方法:取SD大鼠72只,随机分为假手术组、模型组、阳性对照组、NSF组,通过对大鼠实施5/6肾切除手术建立大鼠慢性肾功衰模型;治疗组自二次术后第31 d起给药,连续30 d。检测大鼠血清生化指标Urea、Crea,残余肾组织病理情况及纤维化相关指标:残余肾组织中纤维连结蛋白(FN)、转化生长因子β1(TGF-β1),血清肿瘤坏死因子α(TNF-α)的变化。结果:NSF组与模型组比较:Urea、Crea明显降低(P<0.05),肾脏病理改变较轻,肾组织FN和TGF-β1的表达明显降低(P<0.05),血清TNF-α表达也明显降低(P<0.05)。结论:NSF可能通过下调TNF-α的表达,降低肾脏中纤维化调节因子TGF-β1含量,从而抑制FN增生,控制或延缓肾纤维化进展,发挥对慢性肾功能衰竭的治疗作用。Objeαive: To investigate the mechanisms and effeαs of NSF on prevention of the renal fibrosis in chronic renal failure rats with 5/6 nephreαomy. Methods: 72 SD rats were divided into sham group, model group, positive control group and NSF group, Chronic Renal Failure was induced by nephreαomy. 31 days after the Secondary surgery, treat the positive control group and NSF group for 30 days. Change of Urea, Crea and TNF-α in blood serum were observed and expression of FN and TGF-β protein in nephridial were examined by immunohistochemistry. Results: NSF obviously decreased Urea, Crea ( P 〈 0.05 ), relieved the renal pathological changes and, relieved FN and TGF-β expression in renal tissue (P 〈 0. 05 ) and the expression of TNF-α in blood serum also decreased significantly ( P 〈 0. 05 ) . Conclusion: NSF may inhibit the hyperplasia of FN by decreasing the expression of TNF-α, reducing TGF-β, and interfere renal fibrosis to delay chronic renal failure.
