摘  要:目的建立叶酸偶联载多西紫杉醇壳聚糖(FA-CTS/DTX)纳米粒包封率的测定方法。方法采用高效液相色谱法测定药物含量,色谱柱为Hypersil ODS2,流动相为甲醇-水(67∶33),流速为1.0 ml/min,检测波长为229 nm;通过离心法分离载药纳米粒与游离药物,并进行方法学考察,优化包封率测定条件。结果离心法能有效地将载药纳米粒与游离药物分离,回收率测定结果为101.10%(RSD=2.06%),包封率测定的平均值为85.74%(RSD=0.60%)。结论离心法适用于叶酸偶联多西紫杉醇壳聚糖纳米粒包封率的测定。Objective To establish a HPLC method for the determination of the entrapment efficiency of Docetaxel- loaed, Folate -cojugated chitosan nanoparticles. Method The separation was performed with a Hypersil ODS2 column, and the mobile phase consisted of methanol - water (67:33) with a flow rate of 1.0 mlomin - 1, and UV detection wavelength at 229nm. Centrifugation was applied to isolate free drug for the entrapment efficiency deter- mination. Result Centrifugation could effectively separate the nanoparticles from free drugs. The average recovery was 101.10% ( RSD = 2.06% ), and the average entrapment efficiency was 85.74% ( RSD = 0.60% ). Conclusion The mefilod is simple, accurate and sensitive, and can be used to determine the entrapment efficiency of FA - CTS/DTX nanoparticles.
