摘  要:用紫外线辐射处理野生型龙须菜果孢子体放散刚附着的果孢子,并通过耐高温的筛选,初步获得了2株具有耐高温,生长快速等优良性状的四分孢子体突变株,命名为BZT-11和BZT-18,将其与981和野生型藻株进行了比较分析。结果显示,这2株突变体在实验室内和海上的日平均生长速率为均明显高于981和野生型藻株。在热胁迫时间内,BZT-11和BZT-18的丙二醛(MDA)含量均明显低于野生型,超氧化物歧化酶(SOD)活性明显高于野生型;而在整个胁迫过程中2株突变体与981藻株的MDA含量和SOD活性水平差异不显著。经过4h的热激后,藻株BZT-11和BZT-18的hsp70基因表达水平也显著高于野生型。这些结果进一步证明了该2株突变藻体的耐高温优势。本研究提供了耐高温突变体的筛选技术。The carpospores were mutated by UV irradiation, then thermo-tolerate strains were screened with high temperature. Two mutants of heat tolerance, BZT-11 and BZT-18, were obtained and compared with cultivar 981 and the wild-type strains physiologically. Results demonstrated that the two mutants grew faster significantly than cultivar 981 and the wild-type strains both in the lab and in the sea. The MDA contents in BZT-11 and BZT-18 were all significantly lower than that of the wild during the three days' heat stress. The SOD activities of BZT-11 and BZT-18 were significantly higher than that of the wild, while those mutants and cultivar 981 were at the same level all through the treatment. The hsp70 expressions of two mutants were higher than that of the wild after 4 h heat shock. All these may indicated that the two strains were more tolerant to heat stress. A screening technique for heat-tolerant strains was suggested.
