摘  要:目的建立HPLC—MS/MS法测定Beagle犬血浆中硝苯地平的方法。方法以尼莫地平为内标,血浆样品经乙酸乙酯提取,采用ZorbaxExrend,C-18柱(2.1mm×100mm,3.5μlm)进行分离,乙腈-10mmolNH。CH3COOH缓冲液(印:40)为流动相,流速为0.3m1·min-1,柱温为40℃。质谱采用电喷雾离子化源,正离子检测,扫描方式采用多反应监测(MRM),定量离子分别为硝苯地平m/z347.2→日15.2.内标尼莫地平m/z419.2→日43.3。结果硝苯地平在0.2~200ng-ml。浓度范围内线性关系良好,最低定量限为0.2ng·ml-1。结论本方法简便、准确、专属『生好,适用于硝苯地平在Beagle犬体内血药浓度检测及药代动力学研究。Objective To Beagle dogs. Methods Nimodipine separated on a Zorbax Exrend-Cls develop an HPLC-MS/MS method for the determination of nifedipine in the plasma of was used as the internal standard. After extraction with acetoacetate, nifedipine was colunm(2.1 mm × 100 mm,3.5 μm)at 40 ℃ with acetonitrile-10 mmol anunonium ac- etate(60:4O) as the mobile phase at a flow rate of 0.3 ml -min-1. A tandem mass spectrometer equipped with electros- pray ionization (ESI) source was applied as the detector and operated in the positive ion mode. m/z 347.2→315.2 and m/z 419.2→343.3 were proved to quantify nifedipine and nimodipine respectively. Results The calibration curves were linear over the range of 0.2 - 200 ng ·ml - 1. The lower limit of quantification of nifedipine was 0.2 ng ·ml - 1. Conclusion This method is simple, accurate and suitable for pharmacokinetic studies of Beagle dogs.
