摘　　要：目的建立HPLC—MS／MS法测定Beagle犬血浆中硝苯地平的方法。方法以尼莫地平为内标，血浆样品经乙酸乙酯提取，采用ZorbaxExrend，C-18柱（2．1mm×100mm，3．5μlm）进行分离，乙腈-10mmolNH。CH3COOH缓冲液（印：40）为流动相，流速为0．3m1·min-1，柱温为40℃。质谱采用电喷雾离子化源，正离子检测，扫描方式采用多反应监测（MRM），定量离子分别为硝苯地平m／z347．2→日15．2．内标尼莫地平m／z419．2→日43．3。结果硝苯地平在0．2～200ng-ml。浓度范围内线性关系良好，最低定量限为0．2ng·ml-1。结论本方法简便、准确、专属『生好，适用于硝苯地平在Beagle犬体内血药浓度检测及药代动力学研究。Objective To Beagle dogs. Methods Nimodipine separated on a Zorbax Exrend-Cls develop an HPLC-MS/MS method for the determination of nifedipine in the plasma of was used as the internal standard. After extraction with acetoacetate, nifedipine was colunm（2.1 mm × 100 mm,3.5 μm）at 40 ℃ with acetonitrile-10 mmol anunonium ac- etate（60：4O） as the mobile phase at a flow rate of 0.3 ml -min-1. A tandem mass spectrometer equipped with electros- pray ionization （ESI） source was applied as the detector and operated in the positive ion mode. m/z 347.2→315.2 and m/z 419.2→343.3 were proved to quantify nifedipine and nimodipine respectively. Results The calibration curves were linear over the range of 0.2 - 200 ng ·ml - 1. The lower limit of quantification of nifedipine was 0.2 ng ·ml - 1. Conclusion This method is simple, accurate and suitable for pharmacokinetic studies of Beagle dogs.
