摘  要:目的探讨活性氧(ROS)和不育者精子DNA损伤的关系。方法收集40例精液标本,检测精液中过氧化脂质(LPO)水平及其精子DNA损伤程度。LPO水平以反应产物丙二醛(MDA)的产量表示,测定采用硫代巴比妥酸(TBA)比色法;精子DNA损伤程度采用TUNEL法测定。结果生育组MDA值为(8.243±3.076)μmol/L,不育组为(18.971±4.043)μmol/L,2组相比有极显著性差异(P<0.01);生育组凋亡率(3.68±2.74)%,不育组凋亡率(10.15±4.39)%,2组比较有显著性差异(P<0.01)。结论精子DNA损伤程度与由ROS介导的脂质过氧化反应水平呈正相关。Objective It is to explore the relationship between reactive oxygen species (ROS) and sperm DNA damage in patients with infertility.Methods 40 cases of semen samples were selected and the lipid peroxidation (LPO) level and sperm DNA damage in them were detected.LPO level was indicated by the reaction product of malondialdehyde (MDA) which was measured by thiobarbituric acid (TBA) colorimetric method; sperm DNA damage was observed by TUNEL method.Results MDA value in fertility group was (8.243 ± 3.076) μmol/L,and that in infertile group was (18.971 ± 4.043) μmol/L,there was a significant difference between the two groups (P < 0.01) ; the apoptosis rate in fertility group was (3.68 ± 2.74) % and that in infertile group was (10.15 ± 4.39) %,there was a significant difference between the two groups (P < 0.01).Conclusion Sperm DNA damage was positively correlated with lipid peroxidation mediated by ROS.
