摘  要:目的以天麻块茎为材料,研究组织培养法用于保存新鲜天麻样品对天麻块茎DNA提取的纯度和浓度的影响。方法将新鲜天麻块茎切割成为1cm3形状,接种于MS培养基中,在温度T=21℃的条件下暗培养。分别在培养10d,20d,30d,40d,50d和60d的时候取出样品,再用改良的CTAB法提取其总DNA,测定DNA浓度及纯度。结果组织培养法保存10d,20d,30d后的天麻样品DNA提取,获得的DNA浓度和纯度都较为理想,而保存40d以后提取的样品DNA虽纯度不理想,但浓度较高。结论经过组织培养法保存的天麻样品,能够长时间的保持一定的生长状态,同时也能提取出浓度较高的基因组DNA。Objective To study the effect of the concentration and purity of genomic DNA extraction from Gastrodia elata B1 with the tissue culture method for protecting the tubers of fresh samples. Methods Cut the fresh samples of Gastrodia elata B1 into lcm3, then inoculated them on the MS Medium in dark culture with T=21℃, took out the samples in 10d, 20d, 30d, 40d, 50d and 60d, respectively, obtained DNA by modified CTAB methods, test the concentration and purity of the samples. Results Genomic DNA with high concentration and purity can be got after the 10d, 20d and 30d with the tissue culture method, but high concentration and less purity can be got after 40d on. Conclusion The fresh samples of Gastrodia elata B1 can keep good growth state with the tissue culture method and also can get genomic DNA with high concentration.
