摘  要:目的根据线粒体Cytb基因部分序列,对从入境集装箱中截获的疑似鼠样本进行鉴定种类。方法提取样本中的DNA,采用PCR方法进行线粒体Cytb基因扩增,对目的片段测序,通过blast比对进行同源性比对。从Genbank获取宁波口岸常见8个鼠种24个样本的Cytb基因序列作为参照,利用生物信息学软件Mega5.0进行系统进化分析。结果样本DXR1110与小家鼠同源性最高,DXR1203与褐家鼠同源性最高,系统进化树显示不同鼠种间形成独立分支。结论Cytb基因分析是进行鼠种鉴定的有效手段。Objective To identify species of two suspected samples of rats which found in imported containers based on partial sequences of mitochondrion Cytb gene. Methods Extract DNA from samples, the partial sequences of Cytb gene were amplified using PCR. The target segments were sequenced and sequences alignment had been done through blast. In order to build the phylogenetic tree, 24 cytb sequences were downloaded from Genbank. These sequences belong to 8 species at Ningbo ports. Bioinformatics software Mega 5.0 was applied for phylogenetic analysis. Results The sample of DXRlll0 shared highest sequence homology with Mus musculus and the that of DXRI203 shared highest sequence homology with Rattus norvegicus. Phylogenetic tree show different kinds of rat species forming monophyletic group respectively. Conclusion Analysis of Cytb gene sequences is an effective method to indentify rat species.
