摘 要:目的建立测定痕量大观霉素的共振瑞利散射法。方法在pH9.68的Tris-盐酸缓冲介质中,溴甲酚绿与大观霉素相互作用后共振瑞利散射显著增强,在340nm处,增强的共振光散射强度△IRRS与一定抟度范围的大观霉素线性相关。结果在340nm处,大观霉素的浓度在1.0×10^-7~1.0×10^-6mol/L范围内与△IRRS成正比,方法的检出限(3Sb/s)为0.030mg/L。结论该法用于市售大观霉素药物及尿样中大观霉素含量的测定,结果满意。Objective To develop a novel resonance rayleigh scattering method for determination of trace spectinomycin. Methods In pH9.68 Tris-HCl buffer medium, the resonance rayleigh scattering intensity of bromocresol green was greatly enhanced when spectinomycin was added. And the strongest △IRRS was located at 340nm. The scattering intensity(△IRRs)was directly proportional to the definite quality concentrations of spectinomycin. Results The intensity of RRS was in proportion to the concentration of spectinomycin in the range of 1.0×10^-7~1.0×10^-6mol/L, with a determination limit(3S/S)was 0.030mg/L. Conclusion The method has been applied to the determination ofspectinomycin in the urine and commercially available spectinomycin medicament with satisfactory results.
