摘 要:睡茄素A是存在于催眠睡茄的根和叶中的主要次生代谢产物之一。该实验以催眠睡茄的无菌苗叶片为外植体,利用发根农杆菌C58C1诱导催眠睡茄毛状根,诱导率达30%。同时,利用HPLC测定野生催眠睡茄植株的根、茎、叶和30d液体悬浮培养的毛状根中睡茄素A的含量结果表明,毛状根中睡茄素A平均质量分数为1.588 mg·g-1,是野生植株平均含量的1.96倍;其中,毛状根系M3的睡茄素A含量最高,质量分数达到1.783 mg·g-1,是野生型催眠睡茄植株根含量的1.51倍。因此,可以培养催眠睡茄毛状根来获得睡茄素A。Withanolide A is a biologically active secondary metabolite occuring in roots and leaves of Withania somnifera. In the present study, adventitious roots from leaf explants of W. somnifera were induced for the production of withanolide-A by Agrobacterium tumefacien strain C58C1 to obtain hair roots. Hair roots induction rate reached 30%. The withanolide A was determined by HPLC in different hair roots lines and different parts of W. somnifera. The average content of withanolide A in all hair roots lines were 1.96 times as high as that in wild-plant, the concentration of withanolide A in hair roots ( 1. 783 mg · g^-1 dry weight) were 1.51 times as high as the roots of wild W. somnifera ( 1.180 mg · g^-1dry weight ), respectively. It is possible to obtain withanolide A from hair roots culture of W. somnifera.
