摘  要:目的探讨白细胞介素-1受体拮抗剂(IL-1Ra)及白细胞介素-1β(IL-1β)基因多态性与儿童罹患脓毒症易感性的关系。方法应用聚合酶链反应(PCR)法和限制性片段长度多态性(RFLP)方法,观察30例脓毒症患儿(脓毒症组)和40例健康儿童(健康对照组)的IL-1Ra基因可变串联重复序列(variable number of tandem repeat s,VNTR)多态性及IL-1β基因外显子5多态位点TapⅠ的分布状况。结果脓毒症组IL-1Ra A2等位基因频率和A1/A2基因型分布明显低于健康对照组(P<0.05或P<0.01),而IL-1β基因外显子5多态位点TapⅠ等位基因频率及基因型分布与健康对照组差异无统计学意义(P>0.05)。结论 IL-1Ra A2等位基因在儿童脓毒症易感性中具有保护作用。Objective To explore the association between the interleukinq receptor antagonist (IL-1Ra) and interleukin-1β(IL-1β) gene polymorphisms and sepsis susceptibility in children. Methods The VNTR polymorphism of IL-1Ra gene and polymorphic distribution of exon 5 of IL-1β gene were examined in 30 sepsis children and 40 healthy children using PCR and RFLP a- nalysis. Results Compared with healthy children,A2 allele frequency and A1/A2 genotype distribution of IL-1Ra significantly decreased in sepsis children(P〈0.05 or P〈0.01). There were no significant differences in Tap I allele frequency and genotype distribution of exon 5 of IL-1 β gene between the two groups(P〉0.05). Conclusion IL-1Ra A2 allele may have a protective effect on sepsis susceptibility in children.
