脂联素基因-3971A>G多态性与妊娠糖尿病的相关性研究

摘　　要：目的探讨福建省莆田地区汉族人群脂联素基因(APM1)单核苷酸多态性(SNP)-3971A>G和妊娠糖尿病(GDM)的相关性。方法选择妊娠糖尿病患者108例(GDM组)与糖耐量正常的孕妇112例(对照组),测定其血脂和血糖水平;并采用PCR-RFLP方法鉴定2组APM1基因启动子区-3971A>G的多态性。结果 2组APM1基因SNP-3971基因型与等位基因频率分布比较差异均有统计学意义(P<0.01),GDM组A等位基因频率显著高于对照组(P<0.01)。GDM组A/A基因型患者总胆固醇(TC)和空腹血糖(FBG)水平均高于G/G和A/G基因型(P<0.05),而高密度脂蛋白(HDL-C)均低于G/G和A/G基因型(P<0.05)。结论在莆田地区汉族人群中,与G/G和A/G基因型相比携带A/A基因型者患妊娠糖尿病的危险因素增加。A等位基因可能是妊娠糖尿病的危险因素;APM1-3971A>G与妊娠糖尿病相关。Objective To investigate the association between single nucleotide polymorphism （SNP）-3971A〉G in the promoter region of adiponectin（APM1） gene and gestational diabetes mellitus（GDM） among the Chinese Han population in Putian city, Fujian province. Methods Blood lipid and glucose levels were measured in 108 patients with GDM （GDM group） and 112 pregnant women with normal glucose tolerance （control group）. In addition, -3971A〉G polymorphism in the promoter region of APM1 gene was identified by PCR-RFLP. Results The distribution of allele and genotype frequencies of SNP-3971 was significantly different between GDM group and control group （P〈0.01）. The frequency of A allele in GDM group was higher than that in control group（P〈0.01）. Furthermore, levels of total cholesterol and fasting plasma glucose in patients with A/A genotype were higher than those in patients with A/G or G/G genotype（P〈0.05）. However,levels of high-density lipoprotein cholesterol in patients with A/A genotype were lower than those in patients with A/G or G/G genotype（P〈0.05）. Conclusion Compared with A/G or G/G genotype persons,the risk for GDM increases in A/A genotype persons among the Chinese Han population in Putian. A allele may be a risk factor for GDM and -3971A〉 G polymorphism in promoter region of APM1 gene is associated with GDM.
