摘 要:目的:探讨树突状细胞(DC)在系统性红斑狼疮(SLE)发病机制中的作用以及血必净注射液对SLE 小鼠DC免疫效应的影响.方法:30只SLE模型小鼠(清洁级EB病毒膜抗原BLLF1基因转基因小鼠),均为雌性,体重30~40 g.分为模型组(随机选择8周龄5只和10周龄5只)、8周龄给药组(10只)、10周龄给药组(10只).雌性昆明种小鼠10只作为正常对照组.两给药组连续14 d腹腔内注射血必净注射液6.4 ml/kg后处死,分离脾脏,提取DC和T淋巴细胞,流式细胞术检测DC细胞免疫表型,MTT法检测脾T淋巴细胞增殖活性,并与模型组和正常对照组比较.结果:血必净注射液治疗可以显著升高SLE小鼠脾脏DC表面共刺激分子CD80、CD86和MHC结Ⅱ的表达(P〈0.05或P〈0.01),提高脾脏组织和DC培养上清中IL-12水平,并明显减轻SLE小鼠T淋巴细胞增殖的抑制效应(P〈0.05或P〈0.01).8周龄和10周龄给药组间差异无显著性(P〉0.05).结论:血必净注射液治疗SLE可以改善DC的功能障碍,促进淋巴细胞的增殖和发育.Objective:To investigate the role of dendritic cells (DCs) in pathogenesis of systemic lupus erythematosus (SLE) and the effects of Xuebijing injection (血必净注射液) on immunocompetence of DC in SLE-prone BLLF1 mice. Methods:Ten healthy female Kunming mice aged from 8-10 weeks served as normal control group. Thirty female SLE-prone BLLF1 mice aged 8-10 weeks, were used as the experimental group, and they were di- vided into 3 groups:SLE model group (5 8-week-old and 5 10-week-old BLLF1 mice chosen randomly, n= 10), 8- week-old SLE treatment group (n= 10), and 10-week-old SLE treatment group (n= 10). The two treatment groups received Xuebijing injection 6.4 ml/kg intraperitoneally once a day for 14 days. Spleens were obtained and divided as following: DC were procured by MACS microbeads and T cells by using column of nylon wool. Cells were cultured and phenotypes were assayed by flow cytometry. The proliferation of T lymphocyte of spleen was assessed by MTT. Results:Treatment with Xuebijing injection could significantly up-regulate the expression levels of CD80, CD86 and major histocompatibility complex (MHC) class II of DC, elevate the levels of IL-12 of spleen and supernatant of DC respectively (P〈0.05 or P〈0.01). Meanwhile, Xuebijing injection markedly lowered the inhibitory effect on proliferative activity of T cells in SLE. Conclusions.. Treatment with Xuebijing injection might improve the immune function mediated by DC in SLE, and promote the proliferation of T lymphocyte.
