摘 要:利用双抗夹心酶联免疫吸附法(DAS-ELISA)和反转录一聚合酶链式反应(RT—PCR)对采自云南的胡椒花叶病样品进行检测.结果表明,云南胡椒花叶病的病原为黄瓜花叶病毒(Cucumber mosaic virus,CMV),命名为CMV云南胡椒分离物(CMV—YNP).利用RT-PCR对CMV—YNP外壳蛋白(Coatprotein,CP)基因进行克隆,获得大小为657bp的基因序列.序列分析表明,CMV—YNP与从不同地域感病胡椒中分离的其它CM~株系虽同属CMVIB亚组,但并没有聚在一簇,而且与它们的印基因存在较大差异,核苷酸序列相似性仅在92%~94%.Leaves of black pepper with mosaic symptoms collected from Yunnan province were determined for virus type, and cucumber mosaic virus (CMV) was detected by using the double antibody sandwich enzyme-linked immunosorbent assay and reverse transcription PCR (RT-PCR). CMV isola- ted from infected black pepper was named CMV-YNP. The eDNA fragment of the coat protein (CP) gene of the isolate was obtained by RT-PCR, and it was 657 bp in length. Sequence analysis indicated that CMV-YNP and other CMV strains isolated from black pepper growing in different regions belonged to CMV subgroup IB, hut there were great differences in nucleotide sequence of cp gene. The nucleo-tide identities of cp genes of these CMV strains isolated from black pepper growing in different regions were only 92% to 94%.
