摘 要:目的:研究糖皮质激素性骨质疏松症大鼠骨组织中BMP6/BMP7的mRNA和蛋白表达,揭示糖皮质激素骨质疏松症的发病机理,阐述纳米钙补肾中药的调节作用机制。方法:用糖皮质激素注射的方法复制骨质疏松症模型,用纳米钙及具有益肾填精、补钙壮骨作用的补肾中药高、低剂量对实验大鼠治疗8周,以骨疏康颗粒、盖天力钙片作为阳性对照组,正常大鼠和模型空白组为空白对照组。用PCR法、Western印迹法检测骨质疏松症大鼠骨组织中BMP6/BMP7mRNA和蛋白表达。结果:正常大鼠骨组织可以在基因、蛋白水平表达BMP6及BMP7,模空组大鼠骨组织中的BMP6/BMP7 mRNA和蛋白表达水平明显的下降。纳米钙补肾中药高、低剂量组、盖天力组、骨疏康组,用药8周后,与模空组相比较,均可上调骨组织中BMP6/BMP7 mRNA和蛋白的表达水平。结论:纳米钙补肾中药通过调控大鼠骨组织中BMP6/BMP7 mRNA和蛋白表达,抑制骨吸收,促进骨形成,从而对糖皮质激素性骨质疏松症有一定的防治作用。Objective : To research the bone BMP6/BMP7 mRNA and protein expression of glucocorticoid - induced os- teoporosis rats and to reveal the pathogenesis of glucocorticoid - induced osteoporosis and nanometer - calcium nourishing - kidney herbs. Methods : The osteoporosis model was copied with glucocorticoid injection method. The nano calcium and drugs beneficial for kidney essence and high calcium bones in traditional Chinese medicine were given for 8 weeks with low and high doses. Gushukang Granules and Gaitianli Calcium Tablet were as positive control group. Normal rats and model rats were as the blank group. PCR method and Western blot method were used in the detection of BMP6/ BMPTmRNA and protein expression in osteoporosis rats bone tissue. Results:Normal rats bone tissue can express BMP6 and BMP7 gene and protein level. The model and blank groups' rats bone BMP6/BMP7 mRNA and protein expression levels were significantly declined. Nanometer - calcium nourishing - kidney herbs high and low dose groups, the Gaitianli group and Gushukang group, compared with model and blank groups, all can increase BMP6/BMP7 mRNA and protein expression levels in bone tissue 8 weeks after treatment. Conclusions : Nanometer - calcium nourishing - kidney herbs through regulating the rat bone BMP6/BMP7 mRNA and protein expression, can inhibit bone absorption and promote bone formation, so as to prevent and treat glucocorticoids osteoporosis.
