摘 要:[目的]建立更有效的钻天柳组培快繁技术,以期为其保护及资源利用奠定基础。[方法]以温室内钻天柳带腋芽茎段作为外植体,用正交试验L9(32)研究了不同消毒方法和基本培养基对腋芽萌发的影响。[结果]最佳消毒灭菌方法为:先用70%的酒精消毒10 s,然后用0.1%的氯化汞消毒8 min,此时腋芽污染率最低、腋芽萌发率最高,分别为20.00%、87.50%。极差分析结果表明,酒精处理时间对萌发率影响较大,是诱导腋芽萌发的主要因素;而氯化汞处理时间对污染率影响较大,是影响消毒效果的主要因素。在所试验的5种基本培养基中,腋芽萌发率存在差异,以WPM和ACM培养基效果较佳,萌发率分别达到85.93%和82.59%,与其他处理差异显著(P<0.05);就腋芽的生长状况而言,WPM培养基中芽生长状况最好,叶片呈现深绿色、嫩茎粗壮。[结论]采用70%酒精灭菌10 s,再用0.1%氯化汞消毒8 min后,转入WPM培养基中培养最适宜温室内钻天柳茎段腋芽萌发。[ Objective ] The aim was to set up the high efficient tissue culture and rapid propagation system, and provided theoretical guidance for the conservation and utilization of Chosenia arbutifolia. [ Method ] Axillary buds from stem segments which collected in the greenhouse were used as explant, the impact of disinfection methods, the basic culture medium component on its germination of axillary buds were studied by uhrasound-assist method L9 (32). [ Result] It was found that the time of 70% alcohol disinfection was 10 seconds and the time of 0. 1% HgC12 disinfection was 8 minutes, which the lowest polluting rate was 20.00% and the highest germinating rate of the bud was 87.50%. Range analysis results showed that the effect of time of alcohol treatment on germinating rate was bigger, which was the main factor of inducing germination of axillary buds. But the effect of HgC12 treatment time on polluting rate was bigger, which was the main factor of disinfection effect. In the 5 basic culture mediums, germinating rate of the bud showed some difference. WPM and ACM were the optimal cuhure mediums, their germination on rates were 85.93% and 82.59% , and the difference was significant compared with the other 3 treatments (P 〈 0. 05 ). Growth condition of buds was the best under WPM culture medium condition, whose leaves were deep green and expansion, and stem were stronger. [ Conclusion] The optimum method which was suitable for germination of axillary buds from stem segments of Chosenia arbutifolia was70% alcohol disinfect10 seconds and 0.1% HgC12 disinfect 8 minutes, then inoculate into WPM.
