摘  要:该文建立了HPLC同时测定藏药风毛菊中茵芋苷、东莨菪苷和伞形花内酯3种香豆素类成分含量的方法。采用Wondasil C18-WR色谱柱(4.6 mm×250 mm,5 μm),以甲醇(A)-0.1%磷酸水溶液(B)为流动相,梯度洗脱,流速1.0 mL·min^-1;检测波长325 nm;进样量10 μL;柱温35 ℃。结果茵芋苷,东莨菪苷和伞形花内酯在40 min内均达到基线分离,各成分在其线性范围内均呈现良好的线性关系,线性范围分别为0.18-5.6 μg(r=1.000 0),0.060-1.8 μg(r=0.999 9),0.032-0.97 μg(r=0.999 8)。平均加样回收率分别为99.16%(RSD 0.41%),100.3%(RSD 0.79%),102.2%(RSD 0.87%)。该方法操作简便,准确,重复性和稳定性较好,可作为藏药风毛菊的质量评价提供科学依据。This study is aimed to establish a high-performance liquid chromatography (HPLC) method for simultaneous determination of skimmin, scopolin and umbelliferone in Saussurea hieracioides. Samples were analyzed on a Wondasil C18-WR column (4.6 mm×250 mm, 5 μm) with methanol (A) and water containing 0.1% phosphate (B) as mobile phases for gradient elution at a flow rate of 1.0 mL·min^-1. The detection wavelength and column temperature were set at 325 nm and 35 ℃, respectively, and the sample size was 10 μL. The results showed that skimmin, scopolin and umbelliferone were simultaneously achieved within 40 min under the above conditions. A good linearity was observed in the range of 0.18-5.6 μg (r=1.000 0), 0.060-1.8 μg (r=0.999 9), 0.032-0.97 μg (r=0.999 8) for skimmin, scopolin and umbelliferone, respectively, with the average recoveries of 99.16% (RSD=0.41% ), 100.3% (RSD=0.79%), 102.2% (RSD=0.87%). The method is simple, accurate and reproducible and can be used for the quality control of S. hieracioides.
