摘 要:目的以特性黏数法为对照,采用2种方法测定藻酸双酯钠(PSS)的相对和绝对分子量及其分布,并对2种方法进行比较。方法采用乌氏粘度计测定特性黏数,高效凝胶渗透色谱法(HPGPC)和高效凝胶渗透色谱-多角度激光光散射联用法(HPGPC-MALLS)测定相对和绝对分子量及其分布。结果 2种方法测得的结果存在较大差异,但都与特性黏数法反映相同的趋势。结论与HPGPC法相比,HPGPC-MALLS法测定的结果与真实值更接近,可以用于硫酸多糖类药物的质量控制。Objective To determine the relative and absolute molecular weight as well as distribution of propylene glycol alginate sodium sulfate (PSS) using intrinsic viscosity as control by comparison with two methods. Methods Intrinsic viscosity was determined by ubbelohde viscometer; the relative and ab-solute molecular weight as well as distribution were determined by high performance gel permeation chromatography (HPGPC) and high performance gel permeation chromatography-multiple angle laser light scattering (HPGPC-MALLS). Results The results determined by the two methods showed obvi-ous differences, but they reflected the same trend with intrinsic viscosity method. Conclusion The result of HPGPC-MALLS method is closer to the true values; therefore, the HPGPC-MALLS method can be used for quality control of polysaccharide sulfate drugs.
