摘 要:为检测钙蛋白酶基因(CAPN1)和钙蛋白酶抑制基因(CAST)在牦牛心、肝、脾、肺、肾、胃、眼肌、腹肌等不同组织中的表达水平,采用实时荧光定量PCR技术分析这2个基因的表达规律。结果显示,CAPN1和CAST基因在牦牛的8种组织中均有表达,但在不同组织的表达量存在差异。CAPN1基因在眼肌中的表达量最高,其次为腹肌,在肌肉中的表达量显著高于内脏组织,内脏组织中,在肝脏的表达量最高,其次为肾脏,胃中的表达量最低;CAST基因在胃中的表达量最高,其次为眼肌、腹肌,在肾脏中的表达量最低。This study used real-time fluorescence quantitative PCR to detect the difference expression level of CAPN1 and CAST genes in eight tissues of yak including heart,liver,spleen,lung, kidney,stomach,eye muscle and abdominal. The experimental results showed that CAPN1 and CAST genes were expressed in all of eight tissues of yak,and the expression level of each tissue varied. The highest level of CAPN1 gene expression in eye muscle, and followed in abdominal muscle. The expression level of CAPN1 gene in muscle tissues was significantly higher than other organizations. The highest expression was in the liver,followed by the kidney and the lowest expression in the stomach for visceral tissues. CAST gene expression of the highest level in the stomach,slightly lower in eye muscle and abdominal muscles,and the lowest in kidney.
